Fresh Water Basal
Media
For One Liter
In a 2 liter flask, combine:
980 ml DI Water
0.25g NH4Cl
0.60g NaH2 PO4
0.10g KCL 2.5g NaHCO3
10 ml DL vitamins
10 ml DL minerals
Gas out this media with gas mixture 80% Nitrogen
and 20% Carbon Dioxide over heat while stirring
with a long cannula blowing near the surface
of the media. Once the media approaches a boil,
remove from the heat and cool in an ice bath.
Pipet 9.0 ml into gassed out tubes. Cap, crimp,
and autoclave
